Research data for publication funded by the GREEN-MAP project. A pH-sensitive supramolecular hydrogel based on host-guest interactions between α-cyclodextrin and poly(ethylene glycol) (PEG) chains derived from pH-sensitive micelles was designed for sustained co-delivery of hydrophilic and hydrophobic drugs. The pH-sensitive biodegradable amphiphilic poly(ethylene glycol)-b-polycarbonate was synthesized via ring-opening polymerization of six-membered cyclic carbonate functionalized with a ketal moiety. The amphiphilic copolymer self-organizes into nanoscale micelles able to encapsulate doxorubicin. The host-guest interactions were characterized using X-ray diffraction and differential scanning calorimetry techniques. In vitro release studies revealed that the supramolecular hydrogel exhibited faster drug release profiles in acidic conditions. The MTT assay demonstrated a synergistic cancer cell proliferation inhibition. In vitro cytotoxicity studies indicated excellent biocompatibility of the supramolecular hydrogel matrix.

W publikacji przedstawiono badania dotyczące supramolekularnego hydrożelu opartego na interakcjach gość-gospodarz pomiędzy α-cyklodekstryną i łańcuchami poli(glikolu etylenowego) podchodzącymi od pH-czułych micel, do współdostarczania hydrofilowych i hydrofobowych związków biologicznie aktywnych. Micele wytworzono z diblokowego amfifilowego kopoli(etero-b-węglanu) zawierającego ugrupowanie ketalowe, otrzymanego w wyniku anionowej polimeryzacji cyklicznego węglanu - 9,9-dimetylo-2,4,8,10-tetraoksaspiro[5.5]undekan-3-onu inicjowanej monometoksy-poli(glikolem etylenowym) z wykorzystaniem układu katalitycznego superzasada/tiomocznik. Wprowadzenie ugrupowania ketalowego do struktury kopolimeru pozwoliło na otrzymanie pH-czułych micel, ponieważ zależna od kwasowości środowiska hydroliza tego ugrupowania prowadziła do powstania dwóch grup hydroksylowych osłabiających oddziaływania hydrofobowe w rdzeniu miceli, w efekcie czego, micele pęczniały uwalniając enkapsulowane w micelach i matrycy hydrożelu leki szybciej w kwaśnym środowisku.
