This comprehensive collection contains data related to the development and biological evaluation of cationic bilosome-based nanosystems for targeted treatment of melanoma. To this end, bilosomal carriers were co-loaded with the photosensitizer Rose Bengal (RB) and natural compound astaxanthin (AST) to enhance the efficacy of photodynamic therapy (PDT).
This research was conducted as part of the SONATA BIS 13 project (grant number 2023/50/E/ST4/00603), which is titled "Designing and microfluidic synthesis of hybrid nanocarriers stabilized by novel sugar-based surfactants as a potential “green" anticancer platform”. The project was funded by the National Science Centre in Poland. The study involved detailed physicochemical characteristics and extensive in vitro analysis of human melanoma cell lines (A375 and Me45) and healthy human keratinocytes (HaCaT).
The set of data contains:
1. DLS (Dynamic Light Scattering) and ELS (Electrophoretic Light Scattering) data associated with hydrodynamic diameters, polydispersity index, and zeta potential, including a 30-day storage stability study.
2. Transmission Electron Microscopy (TEM) images confirming the morphology of empty and drug-loaded bilosomes.
3. Singlet Oxygen Generation (SOG) efficiency data, including kinetic calculations and raw spectral measurements using the ABMDMA fluorescent probe.
4. UV-Vis spectroscopic data covering spectral characterization, photobleaching resistance, biological stability in various media, and in vitro drug release profiles at physiological (7.4) and acidic (5.5) pH.
5. Cellular uptake analysis obtained via flow cytometry (quantitative) and fluorescence microscopy (qualitative).
6. Advanced bioimaging data from Holotomographic Microscopy (HTM), providing 2D refractive index maps and 3D volumetric reconstructions of treated cells.
7. In vitro cytotoxicity results (MTT assay) documenting the safety profile (dark cytotoxicity) and phototherapeutic effectiveness (phototoxicity) of the systems.
