Description

This dataset contains experimental data generated to investigate the roles of the transcription factors CIA2 (Chloroplast Import Apparatus 2) and CIL (CIA2-Like) in regulating chloroplast biogenesis during de-etiolation in Arabidopsis thaliana. The dataset includes physiological, molecular, biochemical, and microscopy-based analyses of wild-type (Col-0), cia2, cil, cia2cil, and complemented lines during the transition from darkness to light. Data were generated using chlorophyll fluorescence measurements, pigment analysis, quantitative real-time PCR (qRT-PCR), transmission electron microscopy (TEM), confocal microscopy, luciferase reporter assays, and immunoblot analysis. The results demonstrate that the simultaneous loss of CIA2 and CIL delays chloroplast development, impairs chlorophyll accumulation, disrupts thylakoid membrane organization, reduces photosynthetic performance, and alters the expression of genes involved in chloroplast biogenesis and photosynthesis. Complementation of the cia2cil mutant partially restores these defects, confirming the essential and partially redundant roles of CIA2 and CIL in chloroplast development. This dataset provides a comprehensive resource for investigating the molecular mechanisms underlying chloroplast biogenesis and photomorphogenesis and can be reused for comparative studies of chloroplast development and photosynthetic establishment in plants.
