Synaptic plasticity in the hippocampus is a fundamental mechanism underlying learning and memory. To investigate the temporal dynamics of protein S-palmitoylation during spatial learning, Wistar rats were trained in the Morris water maze using either a short-term (1 h) or long-term (5-day) paradigm. Control groups consisted of naïve cage controls and yoked animals that experienced the same swimming procedures without access to a hidden platform or a stable set of distal spatial cues required for learning the platform location.
Hippocampi were collected immediately after the probe trial and analyzed using acyl-biotin exchange (ABE) coupled with tandem mass tag (TMT)-based quantitative proteomics and LC-MS/MS. The dataset comprises quantitative measurements of protein S-palmitoylation across all experimental groups, identifies differentially palmitoylated proteins (DPPs), and provides site-specific annotation of modified cysteine residues. These data offer a comprehensive, time-resolved view of the hippocampal palmitoylome during spatial learning and constitute a resource for investigating the role of protein S-palmitoylation in synaptic plasticity, learning, and memory.
