This study aimed to synthesize chlorpromazine (CPZ) conjugates with gold nanoparticles (AuNPs) and assess their biological effects on human lymphocyte cell lines (HUT-78 and COLO 720L), as well as human (COLO 679) and murine (B16-F0) melanoma cells. The biological activity of the CPZ-AuNP conjugates was compared to that of free CPZ and well-characterized AuNPs. Cell treatments with CPZ, AuNPs, and CPZ-AuNP conjugates were analyzed using standard assays—MTT, LDH, and MDA—to monitor mitochondrial activity, membrane integrity, and lipid peroxidation, respectively. Results showed that positively charged CPZ-AuNP conjugates were more effective in reducing cell viability than AuNPs alone. A dose-dependent increase in membrane damage was linked to oxidative stress triggered by the conjugates. The biological effects were influenced by both the composition and particle size of the AuNPs. Overall, conjugation with AuNPs appeared to diminish CPZ's biological activity, and the cellular response varied depending on the cell line examined.
